ABSTRACT

@#[摘 要] 目的:探讨甲基转移酶样蛋白7B(METTL7B)在胶质瘤组织中的表达及其与患者临床病理特征和预后的相关性。方法:基于CGGA数据库胶质瘤数据和GTEx数据库正常脑组织数据,分析METTL7B基因在胶质瘤与正常脑组织中的表达差异,并用GEPIA数据库数据和免疫组织化学染色法进行验证。用Kaplan-Meier生存分析、单因素Cox分析、多因素Cox分析及ROC曲线分析等评估METTL7B在胶质瘤患者预后中的价值,用CGGA数据库数据分析METTL7B表达与胶质瘤患者临床病理特征的相关性,用CIBERSORT及TIMER数据库进行肿瘤免疫细胞浸润分析,进行KEGG通路富集分析及GO功能富集分析,通过基因共表达分析确定与METTL7B相关的基因。结果:METTL7B在胶质瘤组织中明显上调(均P<0.05),METTL7B表达是胶质瘤患者独立的不良预后因素。METTL7B高表达与高龄(>41岁)、肿瘤分级增加、肿瘤复发或继发性肿瘤、IDH野生型、1p19q非共缺失以及肿瘤的恶性病理学有关联(均P<0.01);METTL7B表达与B细胞、CD4+ T细胞、CD8+ T细胞、单核细胞、中性粒细胞、巨噬细胞、活化肥大细胞等免疫细胞有关联(均P<0.05)。KEGG通路富集及GO功能分析结果显示,肿瘤相关信号通路及多种免疫反应在METTL7B高表达表型中显著富集。基因共表达分析结果表明,METTL7B与TNFRSF12A、CHI3L1、EMP3表达呈正相关(r=0.807、0.804、0.783,均P<0.01),与ELFN2、REPS2、SHANK2表达呈负相关(r=-0.642、-0.627、-0.602,均P<0.01)。结论:METTL7B在胶质瘤组织中的表达上调是预后不良的指标,且与肿瘤免疫浸润相关。

ABSTRACT

To investigate the clinicopathological and radiological features of benign fibro-osseous lesion (BFOL). Sixty-five cases of craniofacial BFOL, eight cases of peripheral ossifying fibroma (POF) and one case of low-grade central osteosarcoma diagnosed at Sichuan Provincial People's Hospital between January 2010 and March 2019 were collected. The clinicopathologic features, hematoxylin-eosin and immunohistochemical (IHC) staining and radiographic features were analyzed. MDM2 gene amplification was detected by FISH in difficult borderline cases. This cohort of BFOLs included 50 cases of fibrous dysplasia (FD), 12 cases of ossifying fibroma (OF), and three cases of juvenile psammomatoid ossifying fibroma (JPOF). The average ages of patients with FD,OF and JPOF were 31.7, 39.2 and 26.0 years respectively. The male to female ratio was 1.0∶1.8.The average age of POF was 47.0 years, with male to female ratio of 1∶7. Patient of low-grade central osteosarcoma was a 48-year-old man. Twenty-seven cases of FD were located in the jaw, and 23 cases were in other craniofacial bones. Nine cases of OF were located in the jaw, and three cases were in the nasal cavity. Two cases of JPOF were in the nasal sinus, and one was in the jaw. All POF were located in the gingiva, and low-grade central osteosarcoma was located in the mandible. The imaging features of FD were luffa-like or ground-glass like signal shadows with poorly defined borders with expansion. OF had clear borders or sclerosing margins. Both JOF and low-grade central osteosarcoma were expansile intraosseously and with focally invasive nodular masses with ground-glass like signal shadows; and POF showed soft tissue mass with bone formation. Histological features of BFOLs showed mixed fibrous and irregular osteoid lesions. FD had no clear relationship with the host bone and no osteoblasts surrounded the bone trabeculae. Osteoblasts rimming was found in OF, and the boundaries of the host bone were clear. JPOF and low-grade central osteosarcoma infiltrated the host bone focally, and the latter showed mild cellular atypia. MDM2 amplification was detected in low-grade central osteosarcoma. BFOLs are a group of fibro-osseous lesions with similar morphology in the head and neck and face, but their clinical features and prognosis are different; and their imaging and histological characteristics are also slightly different. Attentions should be given to the combination of clinical, imaging and pathologic features of BFOLs, especially the differential diagnosis between BFOLs and low-grade central osteosarcoma. Molecular detection could be used to assist the diagnosis in difficult cases.

ABSTRACT

Background: Hyalodendriella sp. Ponipodef12, an endophytic fungus from a poplar hybrid, was a high producer of botrallin and TMC-264 with various bioactivities. In this study, the influences of eight metal ions (i.e.,Mn2+,Na+, Mg2+,Zn2+,Cu2+,Fe2+,Fe3+ and Al3+) on botrallin and TMC-264 production in liquid culture of the endophytic fungus Hyalodendriella sp. Ponipodef12 were investigated. Results: Three most effective metal ions (Zn2+,Cu2+ and Mg2+) along with their optimum concentrations were screened. The optimum addition time and concentrations of Zn2+,Cu2+ and Mg2+ were further obtained respectively for improving botrallin and TMC-264 production. The combination effects of Zn2+,Cu2+ and Mg2+ on the production of botrallin and TMC-264 by employing statistical method based on the central composite design (CCD) and response surface methodology (RSM) were evaluated, and two quadratic predictive models were developed for botrallin and TMC-264 production. The yields of botrallin and TMC-264, which were predicted as 144.12 mg/L and 36.04 mg/L respectively, were validated to be 146.51 mg/L and 36.63 mg/L accordingly with the optimum concentrations of Zn2+ at 0.81 mmol/L, Cu2+ at 0.20 mmol/L, and Mg2+ at 0.13 mmol/L in medium. Conclusion: The results indicated that the enhancement of botrallin and TMC-264 accumulation in liquid culture of the endophytic fungus Hyalodendriella sp. Ponipodef12 by the metal ions and their combination should be an effective strategy.

ABSTRACT

<p><b>OBJECTIVE</b>To further study the therapy of wasting muscle by myostatin as a new targets, the eucaryotic expression vector coupled the foreign T-helper epitope of tetanus toxin (TT) to the N terminus of myostatin was constructed, and the effects of the gene vaccine on forelimb grip were tested in immunized mice.</p><p><b>METHODS</b>A DNA fragment encoding the TT epitope followed by the N terminus of mature myostatin (330bp) was synthesized. The eucaryotic expression vector of myostatin was constructed and the chinese hamster ovary (CHO) cells were infected with the recombinant plasmids pVAC-TT-Ms by liposome transfection according to routine laboratory procedure. The myostatin expression was tested by cell immunofluorescence technique in transfected CHO. The forelimbs grip were tested in immunized mice with myostatin gene vaccine.</p><p><b>RESULTS</b>The eucaryotic expression vector of myostatin coupled TT epitope was constructed successfully through the restriction analysis and sequencing. The recombinant plasmids pVAC-TT-Ms met quality criterion as gene vaccine by analysis OD260/280 and electrophoresis. The myostatin expression was detected obviously in transfected CHO. The forelimb grip in immunized mice had an obvious increase. The average value of forelimb grip of the mice immunized with pVAC-TT-Ms was about 29.88% greater than that of control mice.</p><p><b>CONCLUSION</b>The construction of eucaryotic expression vector of myostatin coupled TT epitope is successful in expression for recombinant human mature peptide of myostatin. The gene vaccine of myostatin meet quality criterion. The immunized mice has an obvious increase in forelimb grip.</p>

ABSTRACT

Swainsonine, a natural indolizidine alkaloid, has been reported to have antitumour effects, and can induce apoptosis in human gastric and lung cancer cells. In the present study, we evaluated the antitumour effects of swainsonine on several oesophageal squamous cell carcinoma cells and investigated relative molecular mechanisms. Swainsonine treatment inhibited the growth of Eca-109, TE-1 and TE-10 cells in a concentration-dependent manner as measured by MTT assay. Morphological observation, DNA laddering detection and flow cytometry analysis demonstrated that swainsonine treatment induced Eca-109 cell apoptosis in vitro. Further results showed that swainsonine treatment up-regulated Bax, downregulated Bcl-2 expression, triggered Bax translocation to mitochondria, destructed mitochondria integrity and activated mitochondria-mediated apoptotic pathway, followed by the release of cytochrome c, which in turn activated caspase-9 and caspase-3, promoted the cleavage of PARP, resulting in Eca-109 cell apoptosis. Moreover, swainsonine treatment inhibited Bcl-2 expression, promoted Bax translocation, cytochrome c release and caspase-3 activation in xenograft tumour cells, resulting in a significant decrease of tumour volume and tumour weight in the swainsoninetreated xenograft mice groups compared with that in the control group. Taken together, this study demonstrated that swainsonine inhibited Eca-109 cells growth through activation of mitochondria-mediated caspase-dependent pathway.

ABSTRACT

To observe the anti-tumor effect of adoptive immunotherapy with CTL induced by PSA-pulsed dendritic cell vaccine against prostate cancer in mice. Methods: Nude mice were s. c. inoculated with LNCaP tumor cells to establish prostate cancer model. CTL induced in vitro by different peptide-pulsed dendritic cells (Non-DC,Ova-DC,Lys-DC and PSA-DC) were used for adoptive immunotherapy of experiment nude model through vena caudalis on d 15 and d 22 after inoculation. The survival of nude mice and the sizes of tumors were observed with d 50 after the initial treatment as the endpoint. Results: The tumor sizes in Non-DC, Ova-DC, Lys-DC and PSA-DC groups on d 30 and d 50 after adoptive immunotherapy were significantly larger than that at d 15 after inoculation (P<0. 01). The tumors sizes in Lys-DC and PSA-DC groups were significantly smaller than those in the Non-DC and Ova-DC groups(P<0. 01). The survival periods of animals in Lys-DC and PSA-DC groups were significantly longer than those in the Non-DC and Ova-DC groups (P<0. 01). Conclusion: Adoptive immunotherapy with CTL induced by DC-based PSA vaccine can inhibit the growth of prostate cancer LNCaP cells in nude mice and increase the survival of the animals.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the clinical effect of post-extension pulling massage in treating lumbar disc herniation.</p><p><b>METHODS</b>From January 2008 to December 2008, 61 patients with lumbar disc herniation, 34 males and 27 females, ranging in age from 17 to 67 years with an average of 42.6 years, were treated with post-extension pulling massage after continued traction for 30 minutes (on alternate days one time, 3 times as a course of treatment). There was bulging type in 9 cases, hernia type in 22, free type in 30. After a course of treatment, the clinical effects were evaluated according to standard of Macnab, the items included pain, lumbar activity, normal work and life of patients.</p><p><b>RESULTS</b>All patients were followed up from 1 to 9 months with an average of 4.6 months. After treatment, the symptoms and signs of patients had obviously improved in above aspects. According to standard of Macnab, 48 cases got excellent result, 10 good, 2 fair, 1 poor.</p><p><b>CONCLUSION</b>The post-extension pulling massage in treating lumbar disc herniation can obtain satisfactory results, which have localized site of action, small compression for vertebral body and can reduce accidental injury.</p>
